Champions Group C preview: AC Milan is adrift

Group quality in Champions League is a zero sum game. It?s like that in all tournaments. If you have a very strong group (like Group D, this year), either the rest of the groups become easier or one, maybe two groups become so easy that the winner?s less likely to be determined by quality than which team can get their act together in time to advance.

And that?s how we get Group C. It?s a collection of teams that have the talent to make the knockout rounds, but with issues surrounding each at the start of group stage, it?s hard to tell which pair is going to be alive come 2013.

M?laga vs. Zenit St. Petersburg

La Rosaleda, Malaga, 2:45 p.m. Eastern

Continuing Tuesday?s theme of teams having never played each other, Zenit makes the 2,580 mile trip southwest to face M?laga, one of three clubs making their Champions League debut. Sitting second in Spain, M?laga no longer seem like the speed bump they were projecting after the sale of Santi Carzola. Instead, they?re exactly what they were last year: a team Manuel Pellegrini?s made tough to score on, even if they often seem in incapable of scoring more than one goal.

M?laga?s Champions League debut introduces another possible problem, one we saw in Borussia Dortmund?s unexpected troubles in last year. The German champions finished last in their group, reminding us experience in Champions League (and balancing it against domestic concerns) might actually matter. The increased travel, lack of familiarity with opponents, fixture congestion and increased on-field demands are issues clubs often have to confront before they can master. Even clubs that bulk import players and coaches with Champions League experience face trouble. Like Dortmund, Manchester City failed to make last year?s Round of 16.

Zenit did qualify for the knockout rounds, though based on the new blood they?ve brought in, expectations may be higher. Brazilian goal scorer Hulk and Belgian midfielder Alex Witsel made high-priced moves to St. Petersburg earlier this month, right around the time Luciano Spalletti?s team was dropping out of first in the Russia?s Premier League. After a perfect 4-0-0 (W-L-D) start, Zenit?s gone 1-2-1, losing at home to Rubin Kazan and Terek Grozny.

Travelling with only three days rest and having never won in Spain, Zenit?s list of problems creates a matchup of a more talent but travel weary, out-of-form side against tournament neophytes who have the luxury of playing at home. Maybe this one is destined to draw.

AC Milan vs. Anderlecht

San Siro, Milan, 2:45 p.m. Eastern

Until proven otherwise, Milan are Group C favorites. Unfortunately, the Rossoneri seem to be making their opponents? case. They?ve already lost at home to Atalanta and Sampdoria, with only a Giampaolo Pazzini hat trick at Bologna keeping Max Allegri?s side from a completely disastrous start. After selling Thiago Silva and Zlatan Ibrahimovic this summer (and letting a number of veterans go), Milan was supposed to take a step back, but with an easy schedule to start their Serie A campaign, more was expected in the season?s first month.

If Milan lose to Anderlecht, the club will be in full crisis mode, particularly considering the Belgians? history on the peninsula. The Jupiler League champions have made 14 trips to Italy and never won, and while the Italians? struggles make this as good a time as ever to make history, Anderlecht has had their own problems in Belgium. Though they sit second in league, Anderlecht?s won only three of their seven league matches.

Unfortunately for the Belgian Champion (already without ?injured Argentine attacker Mat?as Suarez), Serbian Milan Jovanovic is suspended, leaving the Purple and Whites without two of their four main scoring options from last season. At the rate Milan?s scoring, that may not be an issue, but if Anderlecht?s going to make history instead of merely drawing, they?ll likely need Dieduonn? Mbokani to maintain his form (seven all competition goals this season) or Guillaume Gillet (14 goals last year) to regain his scoring touch.

CHICAGO (AP) - Kevin Correia figured the hitters would be anxious after waiting around for nearly four hours.

He was right.

Correia pitched seven innings of two-hit ball and Starling Marte hit a two-run triple to lead the Pittsburgh Pirates past the Chicago Cubs 3-0 Monday night in a game that was delayed more than 3 1/2 hours by rain.

With the Pirates still in playoff contention, the teams waited out two storms at Wrigley Field that pushed back the start for 3 hours, 37 minutes. The first pitch was thrown at 10:42 p.m. CDT, and Joel Hanrahan delivered the final one at 1:28 a.m. on Tuesday.

"In a game that starts at 10:40, the hitters are going to be aggressive," Correia said. "It's not going to be a game where you take a lot of pitches, so if you can pound the zone with quality pitches, you're going to have a decent outing usually."

Pittsburgh tied Milwaukee for third place in the NL Central at 74-72. Both teams are 2 1/2 games behind division rival St. Louis for the second NL wild-card spot, with the Los Angeles Dodgers also in their path.

Correia (11-9) walked one and struck out six, equaling a season high. He whiffed the first four batters he faced and matched his longest outing of the season.

"You can argue this is the best game he's pitched all year - the length, the efficiency, a couple double plays, a couple minor jams he worked right through and out of," Pirates manager Clint Hurdle said. "Very good command. It was the start we needed, a very good outing by Kevin."

Jason Grilli pitched a hitless eighth and Hanrahan got three outs for his 36th save in 39 chances. Darwin Barney had both of Chicago's hits.

"You can chalk it up to maybe sitting around all day waiting," Cubs manager Dale Sveum said. "Both teams it seemed were in a daze out there."

The Pirates touched up Cubs starter Travis Wood for three runs in the third inning, including Marte's key triple. Jose Tabata drove in Marte with a double to deep center.

Clint Barmes was hit by a pitch and Brock Holt's single up the middle on a 3-2 count sent Barmes to third. After Correia was called out on strikes, Marte tripled to left.

The rookie has tripled in three consecutive games.

"They come in bunches," Hurdle said. "I think the young man has an opportunity to possibly lead the league in triples someday. That's just a lot of usable speed out there. That's the biggest swing of the night for us."

Wood (6-12) worked 5 2-3 innings, giving up four hits. He walked one and struck out a career-high nine.

Hurdle said his team stayed relaxed during the rain delay by watching videos and the Broncos-Falcons NFL game.

"We had plenty of time on our hands," Hurdle said. "We made good use of it.

"Most of these guys know, you're playing 20 (games) in 20 (days). There's no days off. They're not going to reschedule unless the weather just goes way bad. You're here for a while. Professionally, they just settled in and just waited to play."

Pittsburgh loaded the bases with two outs in the sixth, but reliever Michael Bowden retired Holt on a grounder.

Trying to play spoiler and avoid a 100-loss season, the Cubs (58-89) had won seven of eight.

"These guys started talking about it a couple of weeks ago, about (not) losing 100 games. That's their motivating factor, as well as all of us, because nobody wants to be associated with that," Sveum said before the game. "To be able to sit back and knock people out of the playoffs or knock them one game less, that's (also) what you get motivated (for) in September when you're out of it."

1 year on, Occupy is in disarray; spirit lives on

NEW YORK (AP) ? Occupy Wall Street began to disintegrate in rapid fashion last winter, when the weekly meetings in New York City devolved into a spectacle of fistfights and vicious arguments.

Punches were thrown and objects were hurled at moderators' heads. Protesters accused each other of being patriarchal and racist and domineering. Nobody could agree on anything and nobody was in charge. The moderators went on strike and refused to show up, followed in quick succession by the people who kept meeting minutes. And then the meetings stopped altogether.

In the city where the movement was born, Occupy was falling apart.

"We weren't talking about real things at that point," says Pete Dutro, a tattoo artist who used to manage Occupy's finances but became disillusioned by the infighting and walked away months ago. "We were talking about each other."

The trouble with Occupy Wall Street, a year after it bloomed in a granite park in lower Manhattan and spread across the globe, is that nobody really knows what it is anymore. To say whether Occupy was a success or a failure depends on how you define it.

Occupy is a network. Occupy is a metaphor. Occupy is still alive. Occupy is dead. Occupy is the spirit of revolution, a lost cause, a dream deferred.

"I would say that Occupy today is a brand that represents movements for social and economic justice," says Jason Amadi, a 28-year-old protester who now lives in Philadelphia. "And that many people are using this brand for the quest of bettering this world."

On Monday, a couple hundred protesters converged near the New York Stock Exchange to celebrate Occupy's anniversary, marking the day they began camping out in Zuccotti Park.

A handful were arrested after sitting on the sidewalk, but there was no sign of a planned "people's wall" on the streets surrounding the stock exchange.

Instead, protesters held a small meeting where they talked about the ills of Wall Street and corporate greed.

Marches and rallies in more than 30 cities around the world will commemorate the day.

About 300 people observing the anniversary marched Saturday. At least a dozen were arrested, mostly on charges of disorderly conduct, police said.

But the movement is now a shadow of its mighty infancy, when a group of young people harnessed the power of a disillusioned nation and took to the streets chanting about corporate greed and inequality.

Back then it was a rallying cry, a force to be reckoned with. But as the encampments were broken up and protesters lost a gathering place, Occupy in turn lost its ability to organize.

The movement had grown too large too quickly. Without leaders or specific demands, what started as a protest against income inequality turned into an amorphous protest against everything wrong with the world.

"We were there to occupy Wall Street," Dutro says. "Not to talk about every social ill that we have."

The community that took shape in Zuccotti Park still exists, albeit in a far less cohesive form. Occupiers mostly keep in touch online through a smattering of websites and social networks. There are occasional conference calls and Occupy-affiliated newsletters. Meetings are generally only convened to organize around specific events, like the much-hyped May Day event that ultimately fizzled last spring.

The movement's remaining $85,000 in assets were frozen, though fundraising continues.

"The meetings kind of collapsed under their own weight," explains Marisa Holmes, a 26-year-old protester among the core organizers who helped Occupy rise up last fall. "They became overly concerned with financial decisions. They became bureaucratic."

In other words, they became a combustible microcosm of the society that Occupiers had decided to abandon ? a new, equally flawed society with its own set of miniature hierarchies and toxic relationships. Even before the ouster at Zuccotti Park, the movement had been plagued with noise and sanitary problems, an inability to make decisions and a widening rift between the park's full-time residents and the movement's power players, most of whom no longer lived in the park.

"We've always said that we want a new society," Holmes says. "We're not asking anything of Wall Street. We don't expect anything in return."

Occupy organizers in other U.S. cities have also scattered to the winds in recent months. In Oakland, a metal fence surrounds the City Hall lawn that was the hub of protesters' infamous tear-gassed, riotous clashes with police. The encampment is gone, as are the thousands who ventured west to help repeatedly shut down one of the nation's largest ports.

"I don't think Occupy itself has an enormous future," says Dr. Mark Naison, a professor at Fordham University in New York City. "I think that movements energized by Occupy have an enormous future."

Across the nation, there have been protests organized in the name of ending foreclosure, racial inequality, stop and frisk, debt: You name it, Occupy has claimed it. Occupy the Bronx. Occupy the Department of Education. Occupy the Hood. Occupy the Hamptons.

Protesters opposing everything from liquor sales in Whiteclay, Neb., to illegal immigration in Birmingham, Ala., have used Occupy as a weapon to fight for their own causes. In Russia, opposition activists protesting President Vladimir Putin's re-election to a third term have held a series of Occupy-style protests. Young "indignados" in Spain are joining unions and public servants to rally against higher taxes and cuts to public education and health care.

"All around the world, that youthful spirit of revolt is alive and well," says Kalle Lasn, co-founder of Adbusters, the Canadian magazine that helped ignite the movement.

In New York, groups of friends who call themselves "affinity groups" still gather at each other's apartments for dinner to talk about the future of Occupy. A few weeks ago, about 50 Occupiers gathered in a basement near Union Square to plan the anniversary.

There were the usual flare-ups, with people speaking out of order and heckling the moderators. The group could not agree on whether to allow a journalist to take photographs. An older man hijacked the meeting for nearly 15 minutes with a long-winded rant about the NYPD's stop-and-frisk tactics.

A document called "The Community Agreement of Occupy Wall Street" was distributed that, among other outdated encampment-era rules, exhorted Occupiers not to touch each other's personal belongings and laid out rules about sleeping arrangements.

It is this sort of inward-facing thinking ? the focus on Occupiers, not the world they're trying to remake ? that saddens ex-protesters like Dutro, who wanted to stay focused on taking down Wall Street.

Hanging in the entryway to his Brooklyn apartment, like a relic of the past, is the first poster he ever brought down to Zuccotti Park. In black and gold lettering, painted on a piece of cardboard, the sign says: "Nobody got rich on their own. Wall St. thinks U-R-A-SUCKER."

He keeps it there as a reminder of what Occupy is really fighting for. Because despite his many frustrations, Dutro hasn't been able to stamp the Occupy anger out of his soul. Not yet.

On Sept. 17, he'll be down at Liberty Square again. And he'll be waiting, like the rest of the world, to see what happens next.

"We came into the park and had this really magical experience," he says. "It was a big conversation. It was where we all got to realize: 'I'm not alone.'"

Suddenly Summer is over. The roads are busier again as children return to school. For many people their holidays are behind them, the nights are noticeably longer and thoughts turn to Christmas. Let?s look at ways to ensure that Autumn is a time to enjoy.

Let?s look at some tips that can enhance Autumn and make it a pleasant and enjoyable time:

- Make your home cosy. Use luscious, warming fabrics, lovely to touch with bright colours like red, gold, auburn. These will make your rooms welcoming in the evenings. Candle light can really enhance a room, especially scented candles and perfumed diffusers.

- Look at your music collection. How often do you play your older CD?s? Maybe refresh your memory and enjoy working your way through some of your back catalogue. Take a trip down memory lane.

- Locate your slow cooker. It?s lovely to come home after a hard day?s work to the aroma of a meal already prepared and ready to eat. A little organisation with food shopping and menus can mean that you eat healthier and have a more relaxed evening.

- At weekends prepare a stew or casserole. Whilst cooking put on warm clothing and go for a brisk walk with family or friends. You could enjoy a warm bath, a lovely meal and perhaps a film as you settle down upon your return.

- Buy fresh flowers for somewhere unexpected like the landing or bathroom. They?re a treat and will brighten up your home.

- Prepare your garden for Autumn. Winter flowering plants bring colour so that you have a pleasant view from your window. Start feeding the birds and encourage them to visit. It?s lovely to enjoy the many different birds as they learn to come to feed.

- Use your bath. Many of us use the shower for speed but why not relax on a dark evening in a lovely scented bath. Light fragrant candles, have a mug of hot chocolate and allow some time to de-stress and pamper yourself.

- Invite friends round for dinner. If everyone brings a dish it shares the workload, brings fun and enjoyment to your home whilst having an inexpensive evening together.

- Get out your board games. They may be hidden away in a cupboard, but can provide hours of fun and a more sociable evening together than sitting watching television. Record your favourite programmes for another time.

- Children often spend many happy hours absorbed in painting and crafts. They could perhaps start making Christmas cards. Or arrange an indoor picnic and let them help with the preparation. It?s fun to sit on a checked tablecloth in the lounge or kitchen, eating sandwiches and drinking juice.These are the special memories of the future.

Michael Bloomberg has been mayor of New York City since 2002. While he has implemented changes in all areas of life for New Yorkers, his policies concerning health have caused the most controversy. Here are some of his most memorable health proposals, not all of which were enacted: New York City?s Board of Health voted to ban the sale of sugary drinks in containers larger than 16 ounces in restaurants and other venues, in a move meant to combat obesity and encourage residents to live healthier lifestyles. Before the big sugary drink ban proposal, Bloomberg in 2010 urged the U.S. Department of Agriculture to exclude soda, sports drinks and other sugary drinks from food stamp eligibility, citing their effects on obesity. The mayor wanted the food stamp-eligible products to provide nourishment for better health. The department declined, saying the restrictions on retailers were too difficult. Beginning May 23, 2011, smokers were no longer allowed to light up in New York?s parks. Bloomberg proposed the plan in September 2010 to ban outdoor smoking in parks, beaches, marinas, boardwalks and pedestrian plazas, because of secondhand smoke?s harmful effects. Now, it is illegal to smoke in any of the city?s 1,700 parks and open spaces. The punishment is a $ 50 fine. New York?s comprehensive health policy, announced in 2004, included Bloomberg?s goal to help decrease underage binge drinking and illegal alcohol sales through public service ads. After launching a campaign in 2010 to emphasize the hazards of excessive drinking, Bloomberg plans to continue limiting alcohol advertising near schools and to work with the police and the New York State Liquor Authority to enforce laws prohibiting alcohol sales to youths. Bloomberg announced in December that obesity rates among New York public elementary and middle school students decreased over the past five years. He also promoted the Salads in Schools initiative, which provided low-height salad bars to elementary schools across the city?s five boroughs. In October 2007, Bloomberg introduced an initiative for chain restaurants to display calorie information on menus and menu boards. McDonald?s, Burger King and Starbucks previously listed these counts on their websites or posters, but Bloomberg wanted the information to be in plain sight. Counts began appearing on menus, such as this one from Chipotle, in 2008. Salt may liven up meals, but an excess of salt can also lead to some health problems. In January 2010, Bloomberg unveiled a plan to cut the amount of salt in packaged and restaurant food by 25% over a five-year period. In 2006, the New York City Board of Health approved Bloomberg?s plan to ban trans fat in cooking oils within the city?s 24,000 food establishments. The plan gave restaurants 18 months to make the change. New York ? New York City?s Board of Health voted Thursday to ban the sale of sugary drinks in containers larger than 16 ounces in restaurants and other venues, in a move meant to combat obesity and encourage residents to live healthier lifestyles.

The board voted eight in favor, with one abstention.

?It?s time to face the facts: obesity is one of America?s most deadly problems, and sugary beverages are a leading cause of it,? said New York Mayor Michael Bloomberg in a statement earlier this month. ?As the size of sugary drinks has grown, so have our waistlines ? and so have diabetes and heart disease.?

But the move is expected to draw further protest from the soda industry and those concerned about government involvement in their personal choices.

Critics, including McDonald?s and Coca-Cola, have assailed the ban as ?misguided? and ?arbitrary,? though Bloomberg has billed it as both a health and fiscal initiative.

New York City spends an estimated $ 4 billion each year on medical care for overweight people, the mayor said in an earlier statement.

One in eight New Yorkers also suffer from diabetes, a disease often linked to obesity, his office noted, calling sugary drinks ?the single largest driver of these alarming increases in obesity.?

About 58% of New York City adults are considered overweight or obese, the mayor added.

In 2007, the Bloomberg-appointed health board adopted a regulation that forced restaurants to all but eliminate the use of partially hydrogenated vegetable oils and spreads, the main sources of trans fats in the U.S. diet.

Thursday?s decision is expected to take effect in six months and be enforced by the city?s regular restaurant inspection team, allowing restaurant owners nine months to adapt to the changes before facing fines.

?6 months from today, our city will be an even healthier place,? Bloomberg tweeted on Thursday.

The ban would not apply to grocery stores.

The U.S. House of Representatives on Thursday approved a stopgap measure to fund the government for six months and eliminate any threat of a government shutdown fight that could damage lawmakers' re-election hopes.

After spending much of the past two years fighting over cutting government spending, the Republican-controlled House voted 329-91 to slightly raise spending from current levels. Votes in favor were evenly split between Democrats and Republicans.

The bill, which heads to the Senate for a vote expected by next week, will be the last piece of substantive legislation passed by Congress before the November 6 presidential and congressional election. It must be signed into law before September 30, when current government funding runs out.

"We have to pass this important bill to maintain the continuity of our government and to prevent its shutdown," said Representative Harold Rogers, the Republican chairman of the House Appropriations Committee.

The spending measure eliminates one task that Congress needs to complete before it wrangles over the year-end "fiscal cliff" of expiring tax cuts, automatic spending cuts, a debt limit increase and other fiscal deadlines.

By keeping the government's agencies and discretionary programs funded through March 27, lawmakers buy themselves some breathing room in the post-election "lame duck" session to tackle more difficult questions - how to avoid $109 billion in automatic budget cuts that start on January 2, and whether to extend some or all of the tax cuts enacted under former President George W Bush, which expire December 31.

The Congressional Budget Office predicts that the U.S. economy will experience another recession next year if these issues are not resolved. Moody's Investors Service warned on Tuesday that the United States could lose its top-tier credit rating if Congress fails to take action to reduce U.S. debt.

Rival ratings agency Standard and Poor's cut its U.S. rating last year after a grueling battle over raising the debt limit brought the government to the brink of a historic debt default.

That standoff resulted in a budget deal that led to the automatic spending cuts, known as a sequester, which were intended as a painful incentive for Congress to come up with $1.2 trillion in additional deficit reduction. That approach failed and a "meat axe" of $109 billion in across-the-board cuts evenly split between domestic spending and military programs is scheduled to fall on January 2.

New plan on defense cuts
House Republicans Tuesday took another stab Tuesday at avoiding the automatic cuts for defense, passing a bill that orders President Barack Obama to submit a plan to protect military spending by October 15 without any revenue increases.

The measure, approved on a party line vote of 223-196, faces certain death in the U.S. Senate and has drawn a veto threat from the White House.

Republicans in the House passed a similar sequester avoidance plan in May. Authored by Representative Paul Ryan, now the Republican vice presidential nominee, it attempted to halt the cuts and protect military budgets by substituting deep cuts to programs that aid the poor, including food stamps and Medicaid healthcare.

Ryan made his first appearance on Capitol Hill on Thursday since being chosen as Mitt Romney's running mate last month, but did not participate in the debate. He voted for both the funding resolution and the sequester plan.

Amped up with election-season rhetoric, Republicans dug into their positions insisting that deficit reduction come from spending cuts alone, while Democrats insisted on some new tax revenues from the wealthy.

"You tell the president that his plan cannot include one penny of revenue for reducing the deficit. In other words, you say the president's plan has got to look like your plan," Democratic Rep. Chris Van Hollen told Republicans.

Tactical retreat
The six-month budget measure passed on Thursday marks a tactical retreat by House Republicans from their demands for deeper spending cuts this year. The so-called continuing resolution funds the government's agencies and discretionary programs -- from defense to education -- at an annual rate of $1.047 trillion, a level set as part of last year's budget deal.

Republicans were seeking a spending level that was $19 billion lower, but backed off to avoid the risk of a nasty shutdown fight that could hurt their party's election prospects in November. Congressional approval ratings plunged to new lows after last year's debt fights.

But the deal assures them that a post-election lame duck Congress won't be able to push through big spending increases.

The measure also provides some increase in funding for specific programs, including federal efforts to combat wildfires, for nuclear weapons modernization and cybersecurity initiatives.

The continuing resolution drew criticism from the conservative Club for Growth business group.

"This CR (continuing resolution) is bad policy simply because it extends big spending programs, layers on extra riders, and provides only short-term funding for the government so that politicians can leave Washington and/or avoid politically sensitive events," the group said in a statement.

What Are the Most Ecofriendly Cat Litter Products on the Market?

It makes sense that environmentally enlightened cat owners would want cat litter made from natural products that will not potentially compromise their health or that of their beloved pets. Many mass market cat litters contain significant amounts of silica dust which has been linked to upper respiratory issues in cats and even humans. Likewise, the chemical fragrances in many cat litters can also be toxic to cats.

Yet another issue is the sodium bentonite clay in ?clumping? cat litters. The fact that this type of clay can swell up to 15 times its original volume when a cat urinates or defecates into it makes it an excellent cat litter substrate, as waste clumps can be scooped out and filled in without changing the entire litter box. But when cats ingest this material it can cause gastrointestinal distress that in some cases can lead to death. Also, the clay commonly used can be derived from environmentally destructive strip mining.

But thanks to increased concern for cats? health and the environment, there are plenty of greener options out there. To wit, Yesterday?s News cat litter is made from recycled newspaper and is reportedly three times more absorbent than clay. It is non-toxic and contains no scented fragrances, but its makers say it is still tough on odors, and is 99.7 percent dust-free. It also comes in recyclable paper packaging.

Today, two years later, CloudFlare CEO and co-founder Matthew Prince took the stage again at Disrupt to give the crowd a status update. Prince said that at Disrupt 2010, the judges had been concerned about whether or not the startup would be able to scale effectively -- something that can be hard to demonstrate in a demo at a startup pitch competition. In spite of the early skepticism, CloudFlare has been able to hit scale pretty quickly. Prince said that CloudFlare is today providing security and speedy rendering for half a trillion pageviews -- not an insignificant number.

How breast cancer spreads

The invasion of cancer cells into the lymph vessels that connect the breast to surrounding lymph nodes is the first step leading to the metastasis, or spread, of cancer throughout the body. Metastasis is the primary cause of breast cancer deaths. Surprisingly little is known about the control of this process and how it might be interrupted to prolong the lives of women with breast cancer. In a study to be reported Sept. 10 in the Proceedings of the National Academy of Sciences Online Early Edition, researchers at Johns Hopkins describe their discovery of how a protein responsible for cell survival in low oxygen can trigger the spread of cancer cells into the lymphatic system in a mouse model of breast cancer.

The researchers knew that like all solid tumor cancers, breast cancer cells can grow so densely that they end up starved for oxygen. To survive, cancer cells trigger the growth of new blood vessels by activating a protein called hypoxia-inducible factor 1, or HIF-1. "We've known that increased levels of HIF-1 are associated with increased tumor vessels and with patient mortality," says Gregg Semenza, M.D., Ph.D., the C. Michael Armstrong Professor of Medicine, director of the vascular program at Hopkins' Institute for Cell Engineering and a member of the McKusick-Nathans Institute of Genetic Medicine. "Now we've found that HIF-1 activity is directly responsible for the spread of breast cancer to the lymph vessels."

Working in mice injected with human breast cancer cells, which when left undisturbed grow into tumors that spread from the breast to the lungs, Semenza's team previously found that interfering with HIF-1 in these mice reduced growth of the primary tumor and prevented metastasis through blood vessels to the lung. "So of course we wanted to see whether blocking HIF-1 could affect lymph node metastasis as well," he says.

In new experiments, they injected mice with human breast cancer cells that were genetically engineered to knock down HIF-1 protein levels and, after 24 days, examined the mouse lymph nodes to see if the human breast cancer cells had spread. They found that compared to mice whose HIF-1 levels were left undisturbed, lymph nodes with knocked-down HIF-1 contained 76 percent fewer human breast cancer cells, supporting the idea that HIF-1 is somehow involved in the spread of breast cancer to lymph nodes.

To better understand how HIF-1 triggers this to happen, Semenza's team then starved human breast cancer cells of oxygen to see which of the genes involved in the growth of lymphatic vessels might respond to HIF-1. They found that the platelet-derived growth factor B gene ?PDGF-B ? was five times more active when oxygen was lacking. A closer look at the DNA sequence around the PDGF-B gene showed regions of DNA known to be recognized and bound by the HIF-1 protein. They tested this in cells and found that, indeed, HIF-1 protein binds to the PDGF-B gene and turns it on.

The team then took a closer look at PDGF-B to find out how it works once the gene is turned on. They found that PDGF-B that is made by breast cancer cells is pumped out of the cell and stimulates the growth of lymph vessels.

Treating the mice with either digoxin, which blocks HIF-1 activity, or imatinib, a cancer drug, reduced tumor size by 78 percent and reduced lymph node metastasis by 94 percent, although the researchers emphasized that more work must be done to determine whether these drugs will be effective in treating breast cancer patients.

"We're very excited by these results, having shown for the first time that HIFs are directly involved in the lymphatic metastasis of breast cancer," says Semenza. "These results provide experimental support for breast cancer clinical trials that target HIF-1 or PDGF-B." The first study of digoxin in women with breast cancer at the Johns Hopkins Oncology Center will begin later this year.
